Overview

ESB-01 is the first canine supplement built on human longevity research and held to equivalent evidentiary standards. ESB-01 combines three clinically studied actives: nicotinamide mononucleotide (NMN), resveratrol, and quercetin to target the root biology of aging. The formulation addresses the four hallmarks of aging in mammals: NAD+ decline, mitochondrial dysfunction, systemic inflammation, and senescent cell accumulation.

Clinical Trials

ESB-01 on Aging Biomarkers and Functional Outcomes

Aging in companion dogs is characterized by progressive declines in NAD+ metabolism, mitochondrial function, and increased systemic inflammation. This 12-month randomized, double-blind, placebo-controlled superiority trial enrolled 152 senior dogs (aged 6+ years, body weight 6 to 35 kg) across 12 veterinary clinics. Dogs were randomized 1:1 to ESB-01 or matched placebo. Primary endpoints were serum NAD+, C-reactive protein (CRP), malondialdehyde (MDA, oxidative stress), and mitochondrial DNA (mtDNA) copy number. Secondary endpoints were owner-rated activity scale (0 to 10 VAS) and Canine Quality of Life (CQOL) scores. Intention-to-treat analysis used mixed-effects models with Bonferroni correction (α=0.05). Trial followed CONSORT guidelines.

No serious adverse events occurred across the 12-month trial period. Mild gastrointestinal upset was reported in 3% of dogs (n=2 ESB-01, n=3 placebo), all of which were transient. Mean compliance was 92%, assessed via owner diaries and returned capsule counts.

Interactions

ESB-01 has no identified contraindications with common veterinary medications. However, as with any nutraceutical, it should not be administered to dogs with active neoplasia, uncontrolled endocrine disease, or dogs currently receiving medications known to affect NAD+ metabolism or systemic inflammation, without prior veterinary consultation.

Can ESB-01 be taken with other supplements?

ESB-01 may be combined with most standard canine supplements (omega-3 fatty acids, joint support, probiotics). There are no known adverse interactions. Combining ESB-01 with other NMN- or NAD+ -precursor products is not recommended, as the clinical rationale for exceeding the studied dose has not been established in dogs.

Is there a maximum dose?

ESB-01 was evaluated at 1 capsule per 15 kg body weight daily. This dosing protocol was associated with no adverse events in the 12-month trial. More is not necessarily better: the clinical benefits observed derive from the dose shown to be efficacious in the controlled trial, not from exceeding it.

Is ESB-01 appropriate for all dogs?

ESB-01 was studied in dogs aged >6 years, body weight 15~35 kg, without active neoplasia or uncontrolled endocrine disease. Dogs outside this profile, including younger dogs, very small breeds, or dogs with significant comorbidities, were not represented in the trial. If you are unsure whether ESB-01 is appropriate for your dog, consult your veterinarian.

Directions for Use

Administer 1 capsule per 15 kg of body weight daily, with food. For dogs weighing 15 to 30 kg, this corresponds to 2 capsules daily.

 

Dose adjustments are indicated for body weight changes of approximately ±20%. ESB-01 is formulated for long-term daily use; clinical outcomes in the trial were observed at 6 and 12 months of continuous supplementation. 

 

Biomarker improvements were evident at 6 months and sustained through 12 months in the randomized controlled trial. Functional outcomes (activity and quality of life) followed a similar trajectory.
